摘 要:本研究利用小干扰RNA技术探讨端粒酶催化亚单位对鸡马立克氏病MDCC-MSB1细胞端粒酶活性的影响。构建以chTERT基因为靶点的3个RNA干扰载体pRNAT-chTERT-siRNA,筛选并鉴定。利用脂质体转染MDCC-MSB1细胞48 h后,TRAP-PCR-ELISA法定量检测细胞端粒酶活性,并进一步筛选出最有效的小干扰RNA载体。结果显示:转染干扰载体的MDCC-MSB1细胞端粒酶活性逐渐降低,其中干扰载体pRNAT-chTERT-II转染后抑制效果最明显(<0.01)。The study was to investigate the inhibitory effects of siRNA of chTERT gene on telomerase activity in MDCC- MSB 1 cells. The artificially designed and synthesized short hairpin RNA of chicken telomerase catalytic subunit (chTERT) was used to construct the expression vector that was subsequently transfected to MDCC-MSB1 cells, then the telomerase activity of these cells detected by polymerase chain reaction (PCR)-based telomeric repeat amplification protocol (TRAP) coupled with enzyme-linked immune sorbent assay (ELISA). Compared to the control group, the telomerase activities in the treatment ceils showed obvious decrease in 48h after transfection. Among which in the ceils transfected by interference vector pRNAT-chTERT-II had the most evident inhibitory effects. The result showed that this interference vector could effective ely inhibit the telomerase activities of MDCC-MSB 1 cells.
