摘  要:目的:为了使已经石蜡包埋的肾穿组织脱蜡后进行电镜观察,关键是选择适当的固定剂。方法:在石蜡包埋前,肾穿组织分别用甲醛、戊二醛及戊二醛、锇酸双固定。根据光镜和电镜观察结果,选择能兼顾光镜和电镜的固定剂。结果:甲醛固定不能有效保存肾穿组织的超微结构,锇酸的固定能有效地保存超微结构,但严重影响了光镜切片的染色;而戊二醛与甲醛相比,较好地保存了肾穿组织的超微结构,同时又不影响光镜切片的染色。结论:在石蜡包埋前,用戊二醛对肾穿组织进行固定处理,使得利用蜡块组织进行电镜诊断成为可能。另外,二甲苯对蜡块组织的脱蜡完全与否,也是此项实验中影响环氧树脂包埋的一个重要因素。Objective:To choose suitable fixative is key before embedding in order that renal biopsies embedded by paraffin is observed with electron microscopy after deparaffin. Methods:The renal biopsies is fixed respectively by methyl aldehyde,glutraldehyde and osmium tetroxide (OsO 4) after glutraldehyde before paraffin embedding and is observed with light microscopy and electron microscopy.According to the results observed and the demand of light microscopy and electron microscopy,a suitable fixative has been chosen. Results:Methyl aldehyde can't preserves efficiently the ultrastructure of the renal biopsies.Osmium tetroxide can preserves efficiently it but affects dyeing of the section for light microscopy.However,glutraldehyde not only preserves it better than methyl aldehyde but also doesn't affect dyeing of the section for light microscopy. Conclusion:The renal biopsies is fixed by glutraldehyde before paraffin embedding,which enable electron microscopy to diagnose glomerulus disease in renal biopsies embedded by paraffin.The other side,whether deparaffin is complete or not with dimethybenzene,which also is an important factor for epoxy resin embedding in this experiment.
