调脂积冲剂对脂肪肝大鼠肝脏PPAR-γmRNA表达的影响

摘　　要：目的:观察调脂积冲剂对高脂饮食诱导的脂肪肝大鼠的防治作用及对肝脏PPAR-γmRNA表达的影响,探讨其可能作用机制。方法:将SD大鼠49只随机分成空白组、模型组、易善复组、调脂积组。上午,正常组给予普通饲料喂养,模型组、易复善组和调脂积组给予高脂乳剂;下午,两个治疗组分别给予对应药物,空白组和模型组给予生活饮用水灌胃。8周末处死实验大鼠。测定肝脏总胆固醇(TC)和甘油三脂(TG)的含量;HE染色观察肝组织病理变化;采用RT-PCR检测大鼠肝脏PPAR-γmRNA的表达。结果:肝组织出现明显的脂肪变和炎性侵润;与模型组相比,调脂积组TG明显降低;模型组大鼠肝组织PPAR-γmRNA表达减少,调脂积组则显著升高。结论:调脂积冲剂能增加肝脏PPAR-γmRNA的表达。Objective： To observe the preventive effect of Tiao Zhi Ji Granules on high fat diet-induced fatty liver in rats and the liver expression of PPAR-γ mRNA,and to explore its possible mechanism.Methods： 49 SD rats were randomly divided into normol control group,model group,Essentiale group,Tiao Zhi Ji group.Morning,the normal control group were given normal diet,model group,Essentiale group and Tiao Zhi Ji group were given high fat emulsion;afternoon,the two treatment groups were given corresponding drug,normol control group and model group were given orally in drinking water.Rats were sacrificed 8 weeks.Determination of liver total cholesterol（TC） and triglyceride（TG） content;HE staining of liver pathology;using RT-PCR detection of liver expression of PPAR-γ mRNA.Results： The marked liver steatosis and inflammatory rape Run;Compared with model group,TG significantly decreased Tiao Zhi Ji Granules;model of liver tissue with decreased expression of PPAR-γ mRNA,Tiao Zhi Ji Granules was significantly higher.Conclusion： Tiao Zhi Ji Granules could increase hepatic lipid PPAR-γ mRNA expression.
