益气活血中药复方对病毒性心肌炎模型大鼠趋化因子CXCL12 SASH1表达的影响

摘　　要：目的:研究益气活血中药复方影响病毒性心肌炎模型大鼠趋化因子CXCL12(CXC chemokine ligand-12)、Sash1(SAM and SH3 domain containing 1 protein)表达的作用机制。方法:通过改良的抑制性消减杂交技术(SSH),克隆了受CVB3攻击的宿主细胞中被中药调控的基因,并通过荧光rt-PCR对上述结果进行验证。结果:趋化因子CXCL12、Sash1在中药组中呈高表达,病毒组中表达减弱或被抑制。结论:益气活血法能够影响受CVB3攻击的宿主细胞趋化因子CXCL12、Sash1表达,阻断心肌肥大进程,防止向心衰进展。Objective： to explore the influence of compound TCM with the function of reinforcing Qi and activating blood to viral myocarditis model CXCL12（CXC chemokine ligand-12）and Sash1（SAM and SH3 domain containing 1 protein）and its expressed mechanism of action.Methods： Through the improved technique of suppression subtractive hybridization,The gene,which has been attacked by CVB3,has been regulated by TCM,and has located in the host cell,is cloned.Through fluorescent light rt-PCR,the above-mentioned effect can be verified.Results： In TCM group,chemotatic factor CXCL12 and Sash1 are highly expressed,the expression in Virus group is weakened or inhibited.Conclusion： the method of reinforcing Qi and activating blood can influence the expression of CVB3-attacked host cell chemotatic factor CXCL12、Sash1,block the process of myocardial hypertrophy,and prevent its development to congestive heart failure.
