摘  要:目的:研究并建立姜黄素结肠定位给药片的处方及制备工艺。方法:以聚乙二醇4000、聚乙二醇6000和泊洛沙姆为载体分别制备固体分散体,筛选出最佳的固体分散体载体和使用量;将固体分散体压成片剂后,再用尤特奇S100进行包衣,得到姜黄素结肠定位片,并测定体外释放度。结果:最佳的固体分散体载体及其使用量为姜黄素∶泊洛沙姆=1∶4,尤特奇S100包衣增重为8%,结肠定位给药片体外释放度为:0.1mol.L-1盐酸中2h时的释放度为3.25%,pH 6.8磷酸缓冲液中2h时的释放度为3.87%,pH 7.6磷酸缓冲液中90m in时的释放度为75.58%。结论:采用本处方和工艺制备的姜黄素结肠定位给药片可达到结肠定位给药的要求。Objective: To establish the prescription and process of curcumin colon-specific tablet.Methods: Curcumin solid dispersion was prepared and optimized using PEG 4000,PEG6000 and Poloxmer as the carrier respectively;the tablet made from solid dispersion was then coated with Eudragit S100 and In-vitro release of the colon-specific tablet was determined.Results: The best ratio of carrier and curcumin of the solid dispersion was curcumin∶poloxmer =1∶4.The coating weight of the tablet was 8%.In-vitro release was 3.25% in 0.1mol/L hydrochloride within 2 hours,3.87% in pH 6.8 phosphate buffer within 2 hours,75.58% in pH 7.6 phosphate buffer within 90 minutes.Conclusion:The prescription and process of curcumin colon-specific tablet established in the study can achieve the requirements of colon-specific delivery.
