摘  要:目的:探讨从黄芪中同时获取黄芪甲苷和多糖的方法,为确立结肠靶向制剂的最佳提取工艺提供依据。方法:以黄芪甲苷为醇提部分含量测定指标,用超高效液相色谱-蒸发光散射检测器进行测定;以黄芪多糖为水提部分含量测定指标,经苯酚-硫酸法显色,用紫外可见分光光度法进行测定,通过综合评分优选最佳提取工艺。结果:黄芪以70%乙醇,8倍量,提取2次,每次1.5h,提取黄芪甲苷,药渣再用6倍水,提取3次,每次2h,提取黄芪多糖。结论:选出的提取工艺稳定、合理、可行。Objective: To research the extraction to obtain both Astragaloside and polysaccharide so as to provide the basis for the best extraction technology of colon targeted drugs delivery.Methods: In the alcohol extraction part,Astragaloside Ⅳ was determined by UPLC-ELSD as the content mensuration index;in the aqueous part,APS was determined by UV-visible spectrophotometer through Phenol-sulfuric acid method as the content mensuration index,then mark the result to optimise the best extraction of the main component of Astragalus.Results: The optimum extracting condition to extract astragaloside was adding 70 % alcohol with 8 folds to reflux and extract for 2 times,1.5 hours each time.Dregs then was extracted by adding 6 times amount of water to reflux and extract for 3 times,2 hours each time to extract APS.Conclusions: The optimized process is stable and highly efficient.
