摘 要:目的:研究青蒿素对紫外线照射后小鼠表皮c-kit和PAR-2蛋白的表达,探讨青蒿素光保护作用的可能机制。方法:30只BALB/C小鼠分为3组,正常对照组,给予生理盐水灌胃;模型对照组及青蒿素组分别给予生理盐水及青蒿素灌胃后进行UVB照射。各组灌胃均为100mg/(kg.d),每日1次,持续2周。UVB照射每次剂量1 J/cm2,每日1次,共2周,累积剂量14 J/cm2,采用免疫组化检测各组小鼠表皮中干细胞因子受体(c-KIT)和蛋白酶激活受体-2(PAR-2)表达量的变化。结果:UVB反复照射后,青蒿素组皮肤中c-kit和PAR-2表达水平均明显低于模型对照组(P<0.05),且c-kit和PAR-2表达水平呈正相关性(P<0.05)。结论:青蒿素可明显抑制UVB反复照射小鼠皮肤中c-kit和PAR-2的表达,提示系统性应用青蒿素对UVB致小鼠皮肤光损伤有保护作用。Objective:To explore effects of artemisinin on ultraviolet B(UVB)-induced Kit-tyrosine kinase receptor(c-KIT) and proteinase-activated receptor-2(PAR-2) protein expression in mice epidermis,and to investigate the possible mechanism of artemisinin in the prevention of ultraviolet damage.Methods: Thirty BALB/C mouse were distributed into three groups.mouse of normal control group were administrated with normal sodium;mouse of model control group and artemisinin group were gavaged with normal sodiumd and artemisinin respectively,then were irradiated with UVB radiation.A dose of 100 mg/kg/d and 1 J/cm2 was given one time daily for 2 weeks.After UVB radiation,the skin specimens were obtained.Immunohistochemistry method was used to detect the c-KIT and PAR-2 protein expression in these specimens.Results:C-KIT and PAR-2 expression levels in the artemisinin group were significantly lower than those in model control group(P0.05;P0.05).c-KIT and PAR-2 expression levels were correlated in the artemisinin group(P0.05).Conclusion:Artemisinin can inhibit UVB-induced skin c-KIT and PAR-2 expression,which indicates that systemic use of artemisinin may prevent the damage of ultraviolet.
