HPLC测定半夏白术天麻汤中天麻素和鸟苷的含量

摘　　要：目的:建立同时测定半夏白术天麻汤中天麻素和鸟苷含量测定的方法。方法:使用Platisil ODS色谱柱(4.6 mm×250 mm,5μm)柱;流动相A(水)-B(乙腈);梯度洗脱0～30 min(2%～5%B);流速0.65 mL·min-1;柱温25℃;检测波长260 nm。结果:天麻素和鸟苷分别在0.007～0.35 g·L-1(r=0.999 9)和0.000 32～0.016 g·L-1(r=0.999 9)内线性关系良好,平均回收率分别为99.7%和100.6%,RSD分别为1.9%和2.0%(n=9)。结论:所建立的方法准确快速,可用于半夏白术天麻汤复方的质量控制。Objective： To establish an HPLC method for the content determination of gastrodine and guanosine in Banxia Baizhu Tianmatang.Method： The Platisil ODS-C18 column was used.The mobile phases consisted of water and acetonitrile with gradient elution.The flow rate was 0.65 mL.min-1 and column temperature was at 25 ℃.The detection wavelength was at 260 nm.Result： Gastrodine and guanosine were linear in the range of 0.007-0.35 g·L-1（r = 0.999 9） and 0.000 32-0.016 g·L-1（r = 0.999 9） respectively.The average recoveries were 99.7% and 100.6% with the RSD 1.9% and 2.0%（n = 9） respectively.Conclusion： The method was accurate and quick,and can be used for quality control of Compound Pinelliae and Atractylodis Macrocephalae and Grastrodiae Decoction.
