摘  要:目的:研究当药提取物对试验性肝损伤的作用。方法:采用0.1%CCl4油溶液(0.01 mL.g-1体重)连续给药7d,D-氨基半乳糖(D-Gal,800 mg.kg-1)连续给药6 d,分别ip造成小鼠急性肝损伤模型;大鼠ipα-萘异硫氰酸酯(ANIT,100 mg.kg-1),连续给药8 d,造成大鼠急性黄疸模型。各组分别ig当药提取物低剂量(46.3 mg.kg-1)、中剂量(92.6 mg.kg-1)、高剂量(185.2 mg.kg-1)。小鼠摘眼球取血,分离血清,测定血清丙氨酸转氨酶(ALT),天冬氨酸转氨酶(AST)活性和总胆红素(TBIL),直接胆红素(DBIL)含量;大鼠测量胆汁流量,股动脉取血,分离血清,测定AST,ALT,总胆红素TBIL,DBIL水平。各组分别取肝脏,做病理切片,观察肝组织病理改变。结果:同模型组比较,当药提取物92.6,185.2 mg.kg-1能够显著抑制CCl4,D-Gal导致小鼠急性肝损伤和ANIT导致大鼠黄疸模型AST,ALT,TBIL,DBIL的升高,能够减轻模型动物肝脏变性、坏死数量和程度,能够增加黄疸模型大鼠的胆汁流量。结论:当药提取物具有一定的保肝作用。Objective:To study the protection effects of Swertia pseudochinensis Hara extract on experimental liver injury.Method: Mouse model of acute liver injury was established by ip injection of 0.1% carbon tetrachloride(CCl4) oil solution(0.01 mL.g-1),and D-Galactosamine(D-Gal,800 mg.kg-1) respectively.Rat model of acute jaundice was developed by ip injection of α-naphthyl isothiocyanate oil solution(ANIT,100 mg.kg-1).Several doses of S.pseudochinensis Hara extract were ig administrated to the models.Activity of alanine aminotransferase(ALT),aspartate aminotransferase(AST) and total bilirubin(TBIL),direct bilirubin(DBIL) Content in serum were determined.Bile flow in rats and pathological changes in liver were also investigated.Result: S.pseudochinensis Hara extract at the concentration of 92.6 mg.kg-1 inhibitted the acute liver injury induced by CCl4 and D-Gal in mice.It decreased the increasing level of transaminases caused by ANIT in rat jaundice model,and reduced the liver degeneration and the number and extent of necrosis in the model animals.The extract also increased the bile flow rate in the rat jaundice model.Conclusion: S.pseudochinensis Hara extract has the efficacy of protecting liver from injury to a certain extent.
