摘 要:目的:观察GBE50对缺氧大鼠肺动脉高压的保护作用。方法:复制常压低氧肺动脉高压大鼠模型。30只健康SD大鼠随机分为正常对照组(N)、模型组(M)和GBE50组。GBE50组于每天缺氧后按照80 mg.kg-1口服GBE50,21天后观察管腔面积/管总面积(LA%)、RT-PCR测肺组织中的iNOS(诱导性NO合酶)活性的变化、用放免法测血浆中cGMP、NO含量。结果:与正常对照组比较,模型组cGMP、LA%明显降低(P<0.01),iNOS mRNA表达明显升高(P<0.01)和NO的含量明显增加(P<0.05)。与模型组比较,GBE50组LA%、iNOS mRNA明显下降(P<0.05)和NO的含量明显增加(P<0.05),cGMP含量差异无统计学意义(P>0.05)。结论:GBE50对慢性间歇性缺氧大鼠肺动脉高压有一定的保护作用。Objective: To investigate the protective effects of Ginkgo Biloba Extract(GBE50) on pulmonary artery hypertension in chronic intermittent hypoxia rats.Methods: Thirty healthy Sprague-Dawley rats were randomly divided into normal group,model group and GBE50 group.The hypoxia pulmonary artery hypertension models were established in model group and GBE50 group.The rats in GBE50 group were administered GBE50 of 80 mg·kg-1 every day.After treatment of 21 days,the vessel cavity area / total area(LA%) was observed and the activity of iNOS in lung tissue and the contents of cGMP and NO in blood plasma were tested by RT-PCR and radioimmunoassay method respectively.Results: The levels of cGMP and LA% decreased and the expression of iNOS mRNA and the content of NO increased in model group obviously(P0.05 or P0.01) compared with normal group.The level of LA% and the expression of iNOS mRNA decreased and the content of NO increased in GBE50 group significantly(P0.05)compared with model group.Conclusion: GBE50 has protective effects on pulmonary artery hypertension in chronic intermittent hypoxia rats.
