摘  要:目的:观察鼻渊舒口服液对兔慢性鼻窦炎(CRS)模型鼻窦黏膜IL-8,TNF-αmRNA表达的影响,探索鼻渊舒口服液治疗CRS的可能机制。方法:取新西兰大白兔60只,适应性喂养1周后,随机分为正常组、模型组、假手术组、鼻渊舒组、西药治疗组,每组12只,建立CRS模型;鼻渊舒组、西药治疗组分别给予鼻渊舒口服液1.5 mL.kg-1d-1,克拉霉素25 mg.kg-1.d-1,共14 d,处死后取鼻窦黏膜组织HE染色光镜观察,实时定量PCR法检测鼻窦黏膜IL-8,TNF-αmRNA表达。结果:模型组鼻窦黏膜呈慢性炎症表现,黏膜炎细胞浸润,上皮细胞、腺体和杯状细胞明显增生;鼻窦黏膜IL-8,TNF-αmRNA表达较正常组显著增高(P<0.01)。经鼻渊舒治疗后鼻窦黏膜上皮修复较好,炎细胞浸润、腺体和杯状细胞增生不明显;鼻窦黏膜IL-8,TNF-αmRNA表达较模型组显著降低(P<0.01),与正常组比较无显著差异。结论:鼻渊舒口服液治疗CRS的机制可能与调控鼻窦黏膜IL-8,TNF-αmRNA表达有关。Objective:To observe IL-8 and TNF-α mRNA expression of rabbit sinus mucosa of Biyuanshu treatment of chronic rhinosinusitis(CRS) model,and explore the possible mechanism of Biyuanshu treatment of CRS.Method: Sixty new Zealand rabbits fed one week after adaptation and were randomLy divided into normal group,model group,sham operation group,Biyuanshu group,western medicine group,12 in each group.Firstly CRS model have established,then Biyuanshu group and western medicine group were given 1.5 mL.kg^-1 of BiYuanShu or 25 mg'kg-1of clarithromyein every day inl4 days, finally they were sacrificed to take sinus mucosa, HE staining were observed by light microscopy and IL-8 and TNF-α mRNA expression of sinus mucosa were detected by real-tlme quantitative PCR. Result: In model group, the sinus mucosa showed chronic inflammation and mueosal infiltration of inflammatory cells, and epithelial cells and glands and hyperplasia of goblet cells were found, IL-8 and TNF-α mRNA expression of sinus mueosal were higher than normal group significantly (P 〈 0.01). In Biyuanshu group, sinus mueosa have good repair, and inflammatory cells and glands and goblet cell hyperplasia were not obvious, IL-8 and TNF-α mRNA expression of sinus mueosal were lower than model group significantly (P 〈 0.01 ), Biyuanshu group and normal group have no significant difference. Conclusion: The mechanism of Biyuanshu treatment of CRS may be related to regulation of IL-8 and TNF-c~ mRNA expression of sinus mucosal.
