摘 要:采用原生质体分离、菌丝尖端分离和组织分离3种方法对双孢蘑菇(Agaricus bisporus)As2796的退化菌株进行分离,应用灰色关联度分析法综合评价不同分离株的农艺性状,筛选综合性状优于As2796退化菌株的提纯复壮菌株。结果表明,3种分离方法均能获得提纯复壮的菌株,其中原生质体分离株P28的提纯复壮效果最好。Protoplast,tissue and hyphal tip isolation methodologies were used to isolate rejuvenated strains from the retrogressive strain Agaricus bisporus As2796.Mycelia growth rates on potato-dextrose-peptone-yeast(PDPY) medium,wheat spawn medium and composted substrate were determined and used to select 27,five and four strains,isolated by protoplast,tissue and hyphal tip isolation methodologies,respectively for fruiting tests.Seven agronomic traits(cap diameter and thickness,stipe diameter and length,average fruit body weight,yield per square meter and period required for substrate colonization) were evaluated by gray correlation analysis.Comparison of weighting coefficient values revealed that isolates P28,Tip8 and TI12 exhibited the most favourable fruit body and cultivation characteristics among the rejuvenated strains obtained using protoplast,tissue and hyphal tip isolation methodologies,respectively.Overall,these strains ranked 1st,7th and 3rd,respectively among the 36 selected rejuvenated isolates.
