摘  要:目的为研究体外培养小鼠神经干细胞(NSC)血小板源性生长因子(PDGF)、表皮生长因子(EGF)和胶质细胞源性神经营养因子(GDNF)的基因表达.方法取小鼠胚胎海马NSC培养并鉴定,提取其总RNA,用RT-PCR技术检测培养NSC内PDGF、EGF、GDNFmRNA的表达.结果培养NSC中PDGF mR-NA表达最强,EGF mRNA明显表达,但几乎未检测到GDNF mRNA.结论为了解NSC分泌神经营养因子发挥生物学作用提供了理论基础.Objective To investigate the expression of platelet derived neurotrophic factor(PDGF),epithelial growth factor(EGF),and glia derived neurotrophic factor(GDNF)in the neural stem cells(NSC) of mice in vitro.Methods We isolated NSCs from hippocampus of mice embryo(9 days)and cultured for 7 days,and then identified(nestin staining)them by using immunohistochemisstry method.Total RNA was extracted from NSCs and the Mrna expression of PDGF,EGF-1,and GDNF was determined by RT-PCR.Result In cultured NSCs in vitro,intensive expression of PDGF mRNA was seen and the EGF mRNA was also detected,while there was no detectable level for GDNF.Conclusion The present study provides some basic evidences to know the expression of PDGF,EGF and GDNF in cultured NSCs,which is available to understand the role of NSCs by secreting neurotrophic factor for their neurotrophic effect.
