摘  要:目的全面了解参与大肠腺瘤癌变的表皮生长因子受体信号通路(epidermal growth factor receptorpathway,EGFR pathway)基因.方法分别提取正常大肠粘膜、大肠腺瘤、DukesA、DukesB和DukesC-D大肠腺癌组织的总RNA,生物素标记cRNA探针,分别与5张Affymetrix U133 PLUS 2.0基因芯片杂交,GeneScanner 3000扫描,GCOS1.2软件处理杂交信号,将大肠腺瘤与大肠腺癌(包括DukesA,DukesB和DukesC-D大肠腺癌)与正常大肠粘膜组相比较筛选表达差异基因.应用PathwayV1.0软件对以上筛选出的表达差异基因进行通路分析.结果 EGFR信号通路参与正常大肠粘膜→腺瘤→腺癌这一过程(P<0.05),EGFR信号通路中共有包括MAPK,Shc1,SOS1、Grb和c-FOS基因等29个基因在大肠腺瘤癌变过程中有显著的表达差异(P<0.05),这些基因在大肠腺瘤癌变过程中发挥着重要作用.结论 EGFR信号通路中共有29个基因参与大肠腺瘤癌变,对选择EGFR及其下游信号转导因子作为抗肿瘤治疗的重要靶点有一定指导意义.Objective To comprehensively understand the epidermal growth factor receptor pathway(EGFR pathway)genes involved in the colorectal adenoma-carcinoma transition.Methods Total RNA was extracted form tissues of normal human colorectal mucosa,colorectal adenoma,DukesA,B and C-D colorectal cancer respectively,biotin-labled cRNA target probes were hybridized with 5 Affymetrix Human U133 PLUS 2.0 Gene Chips.Signal images were scanned by gene scanner 3 000 and analyzed with GCOS1.2 software.Then differentially expressed genes were screened out by compared colorectal adenoma and colorectal adenocarcinoma(including DukesA,DukesB and DukesC-D colorectal adenocarcinoma)with normal colorectal mucosa.Then pathway analysis of differentially expressed genes sets was performed by using Pathway V1.0 software.Results EGFR pathway participated in the process of colorectal adenoma-carcinoma transition(P0.05).There were 29 differentially expressed genes of EGFR pathway,including MAPK,Shc1 SOS1,Grb and c-FOS genes,involved in the process of colorectal adenoma-carcinoma transition(P0.05),these genes played important roles in the carcinogenesis and progression of colorectal carcinoma.Conclusion There are 29 genes of EGFR pathway participate in the colorectal adenoma-carcinoma transition,the results have some guiding significance for selecting EGFR or it's downstream signal transduction factors as anti-tumor therapeutic targets.
