摘 要:采用半叶枯斑法、叶圆盘法测定了鸦胆子素D对黄瓜绿斑驳花叶病毒(CGMMV)的抑制效果,结果表明,20μg.mL-1的鸦胆子素D对CGMMV的侵染和增殖均有较强的抑制活性,其对CGMMV侵染和复制抑制率分别为92.6%和88.3%;实时荧光定量PCR测定结果表明,20μg.mL-1该化合物对CGMMV复制酶基因、运动蛋白基因和外壳蛋白基因表达的抑制率分别为88.8%、92.1%和90.5%.采用Western blot方法测定的结果表明,鸦胆子素D可显著抑制黄瓜叶片中CGMMV CP和MP蛋白的表达.The activity of Brucein D against Cucumber green mottle mosaic virus(CGMMV) was investigated by local lesion and leaf discs assay,the results indicated that the compound showed a significant inhibitory activity against CGMMV in vitro,with 92.6% inhibitory ratio against the infection of CGMMV and 88.3% against the replication of CGMMV at the concentration of 20 μg·mL-1.While its inhibitory effect on expression levels of CGMMV Rep,MP and CP gene in cucumber leaf discs was examined by real-time quantitative PCR,which showed that the compound obviously suppressed the expression of genes,with 88.8%,92.1% and 90.5% inhibitory ratio at the concentration of 20 μg·mL-1,respectively.In addition,Western blot test results revealed that the compound obviously suppressed the expression of CGMMV CP and MP in cucumber leaves.
