摘  要:目的:探讨Integrin介导的ERK信号传导通路在滋养层细胞侵袭过程中的作用.方法:利用人绒毛外细胞滋养层细胞体外侵袭模型,将不同浓度的integrin α1和integrin β1混合抗体分别加入绒毛外细胞滋养层细胞(EVCT)培养液中,培养4 h,观察integrin α1和integrin β1混合抗体对EVCT的生长、体外侵袭、FAK和ERK1/2磷酸化的影响.结果:integrin α1和integrin β1混合抗体呈浓度依赖方式抑制EVCT中FAK磷酸化程度,同样呈浓度依赖方式抑制磷酸化的ERK1/2表达,P〈0.05.结论:细胞外基质(ECM)成分能通过细胞表面的相应受体integrin α1β1,激活滋养层细胞中重要的蛋白激酶FAK和ERK.ERK信号传导通路对滋养层细胞的侵袭行为起到重要的调节作用.Objective:To study the effect of Integrin-mediated ERK signaling pathway on the process of trophocyte invasion. Methods:Extravillous eytotrophoblasts (EVCT) invasion model in vitro was established, and then we put different concentrations of integrin α1 and integrin β1 antibodies into the EVCT medium for 4 h, and observed the effect of integrin α1 and integrin β1 mixed antibodies on growth, in vitro invasion of EVCT, FAK and ERK1 phosphorylation. Resuls:The phosphorylation of FAK in EVCT was dose-dependently inhibited by integrin α1 and integrin β1 combined antibody, and its antibody also dose-dependently inhibited expression of phosphorylated ERK1/2. Conclusions: Extracellular matrix (ECM) may activate FAK and ERK in trophocytes by correspondent integrin α1β1 on cell surface. The ERK signal transduction pathway regulates the invasion of EVCTs.
