摘  要:目的观察饥饿和再进食对小鼠下丘脑弓状核中obestatin表达的影响,探讨下丘脑弓状核中obestatin在摄食调节中的作用。方法 50只♂昆明小鼠随机分为5组:A组为饥饿0h,B组为饥饿24h,C组为饥饿48h,D组为饥饿72h,E组为饥饿72h后再进食4h。采用免疫组化法检测饥饿不同时段和再进食后下丘脑弓状核中obestatin表达的变化。结果 B、C、D、E组弓状核obestatin表达水平均明显低于A组,差异有统计学意义(P<0.05)。与D组比较,E组弓状核obestatin表达略升高,差异无统计学意义(P>0.05),A、B、C、D组弓状核obestatin表达呈逐渐降低趋势,差异无统计学意义(P>0.05)。结论 obestatin在弓状核中高表达,饥饿可减少obestatin在弓状核中的表达,再进食后无明显变化。提示弓状核中obestatin可能通过其表达的减少参与饥饿状态时摄食行为的调节,而对短期再进食状态时的摄食行为无调节作用。Objective To study the effects of fasting and refeeding on the expression of obestatin in the hypothalamic arcuate nucleus(ARC) of mice.The roles of obestatin in ARC on the regulation of food intake were investigated.Methods Fifty Kunming male mice were used in this study.Group A: fasted for 0 h,Group B: fasted for 24 h,Group C: fasted for 48 h,Group D: fasted for 72 h,Group E: refed for 4 h after fasted for 72 h.The expression of obestatin in the ARC on different times of fasting and refeeding were studied by the technique of immunohistochemistry.Results The expression of obestatin in the ARC from B group to E group were significantly lower than those of group A(P 0.05) .Compared with group D,the expression of obestatin in the ARC from group E were slightly increased.The expression of obestatin in the ARC from group A to group D were decreased,but it showed no significant difference.Conclusion Obestatin is highly expressed in the ARC and that fasting decreases the obestatin in the ARC,but this expression do not change after refeeding.Our results indicate that obestatin in the ARC may participate in the regulating of feeding behavior on fasting through the decreasing of its expressions,and have no action on the feeding behavior of short-term refeeding.
