摘 要:目的:探讨力竭运动后中枢神经系统内核转录因子(NF-κB)信号通路的活化和调控机制及与其下游靶基因产物IL-1β介导的炎症反应之间的相互关系。方法:健康雄性SD大鼠70只随机分为对照组(n=10)和力竭组(n=60)。力竭组进行1周力竭游泳训练后,选择即刻、4、8、12、16、24h等不同时间点取脑,经Western-Blot半定量分析研究运动后杏仁体内NF-κB的含量变化规律;免疫组织化学SABC染色法观察IL-1β在杏仁体神经元内的表达变化。结果:NF-κB在力竭4h开始激活(P<0.05),至8h达到高峰(P<0.05),12~16h开始回落(P<0.05),24h恢复到正常对照水平(P>0.05);IL-1β免疫阳性神经元的数目在24h内持续且高于对照(P<0.05),12h达峰值(P<0.05),其余各组之间差异无显著性(P>0.05)。结论:NF-κB信号系统和IL-1β参与了脑内的炎症反应。Objective:To investigate the mechanism of activation and regulation of NF-κB signaling pathway in central nervous system(CNS) and the relationship between NF-κB signaling pathway and the IL-1β induced inflammatory reaction in rats following exhausing exercise.Methods:Seventy healthy SD rats were randomly divided into control group(n=10) and exhausted swimming group(n=60).The brains were dissected at 0,4,8,12,16,24 h after seven-days of exhausting exercise.With the Western Blot technique,the expressional changes of NF-κB in each group were observed and detected;and the expression of IL-1β was detected by immunohistochemical SABC techniques.Results:NF-κB was activated at 4 h(P0.05),peaked at 8 h(P0.05) after exhausting exercise,decreased at 12-16 h(P0.05),and returned to normal levels at 24 h(P0.05).Compared with control group,the number of IL-1β positive cells increased dramatically in all exercise groups in 24 h(P0.05),peaked at 12 h(P0.05),there was not significant difference in other exercise groups(P0.05).Conclusion:The present data suggest that NF-κB signaling pathway and IL-1β are involved in inflammatory reaction of brain.
