摘  要:目的:评价脯氨酸二硫代氨基甲酸酯(PDTC)对大鼠子宫内膜异位症(EMT)模型的治疗效果。方法:将构建成功的EMT模型鼠分为PDTC治疗组(PDTC组),亮丙瑞林(LEU)治疗组(LEU组)及未干预组,未成模鼠作为阴性对照组。测量用药前后各成模组异位内膜体积,并采用酶联免疫分析(ELISA)法比较各组大鼠血清中白细胞介素1β(IL-1β)和肿瘤坏死因子-α(TNF-α)蛋白表达水平;采用免疫组化SP法测量各组中核因子κB(NF-κB)的表达。结果:①治疗后各研究组异位内膜体积均缩小,PDTC组及LEU组与未干预组之间差异均有统计学意义;而PDTC组与LEU组之间差异无统计学意义。②阴性对照组血清TNF-α及IL-1β的表达水平比未干预组低,而PDTC及LEU两干预组的表达均降低。结论:PDTC可使大鼠EMT模型异位病灶显著缩小及血清中的TNF-α及IL-1β表达降低。Objective:To evaluate the treatment effects of pyrrolidine dithiocarbamate (PDTC) on endometriosis(EMS) in rats.Methods:The successfully built model of EMT rats were divided into PDTC group,leuprorelin (LEU) group and untreated group. The failed model was as negative control group. The size of endometrial explants in each group was measured before and after medication. The expression of interleukin-1 (IL-1) and tumor necrosis factor (TNF-α) in each rat serum was measured by enzyme-linked immunosorbent assay (ELISA) and the protein level of nuclear factor (NF-kB) in each group was analyzed by SP immunohistochemical method.Results:①The size of endometrial explant in PDTC and LEU group was significantly decreased compare with untreated group,there was significant difference. While there was no statistical difference between the PDTC and LEU group. ② The expression of TNF-α and IL-1β in negative control group was lower than that in untreated group,while which was decreased in PDTC and LEU group.Conclusions:PDTC can significantly decrease the volume of endometrial explant and lower the expression of IL-1β and TNF-α in EMT rat model.
