摘 要:采用RAPD标记技术,评估中国西南地方优良种质巫溪(W)、美国玉米带坚秆综合种BSSS9(B)、墨西哥热带种质墨黄九(M)及以它们为主体亲本人工合成的WBMC0基础群体、改良一轮的WBMC1群体的遗传变异。结果表明:(1)15个10-mer随机引物共扩增89个DNA片段,其中显示多态性的片段,在W、B、M、WBMC0、WBMC1中分别为68、67、71、76和82个,其多态片段比例依次为76.4%、75.3%、78.9%、85.4%和92.1%;(2)5个群体各89个片段的平均杂合度W为0.285、B为0.252、M为0.296、WBMC0为0.327、WBMC1为0.346,以WBMC0和WBMC1较大;(3)5个群体各89个片段的平均遗传距离W为0.2533、B为0.2246、M为0.2481、WBMC0为0.3006、WBMC1为0.3119,也以WBMC0和WBMC1较大;(4)累加各引物扩增的基因型种类,W为136、B为117、M为128、WBMC0为151、WBMC1为150。以上结论说明,人工合成的广基玉米群体,遗传多样性比亲本群体丰富,且这种多样性属DNA变异,并非环境造成。经过一轮选择后的?The genetic diversity of a maize population Wuxi(W) from Southwest China, BSSS9 (B) from America, Mohuangjiu (M) from Mexico , WBMC0 synthesized with W, B, M as main parents, WBMC1 one cycle selected from WBMC0 was evaluated by RAPD molecular marker.The results showed that (1)totally 89 fragments(loci)were amplified by 15 10mer random primers,the proportion of polymorphic loci were W 76.4%, B 75.3%, M 79.8%, WBMC0 85.4% and WBMC1 92.1% respectively; (2)the mean gene heterozygosity based on 89 loci was W 0.285, B 0.252, M 0.296,WBMC0 0.327 and WBMC1 0.346;(3)the mean genetic distances based on 89 loci were W 0.2533, B 0.2246, M 0.2481, WBMC0 0.3006, WBMC1 0.3119;(4) the genotypic numbers accumulated by 15 primer amplification were W 136,B 117,M 128,WBMC0 151 and WBMC1 150.All indexes indicated that the synthesized maize populations were more polymorphic than the parent populations in DNA level.One cycle selection did not reduce the variation. The principle and technical system were discussed for evaluating genetic variation of reccurent selection population using RAPD molecular marker.
