摘 要:目的:研究马鞭草科紫珠属植物裸花紫珠Callicarpa nudiflora的化学成分,并筛选其止血活性成分。方法:应用硅胶及Sephadex LH-20柱色谱方法分离纯化,运用波谱数据解析鉴定化合物结构;测定分离得到的化合物对家兔体外血液凝集参数凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)及纤维蛋白原(FIB)含量的影响。结果:分离鉴定了11个化合物,其中2个二萜:7α-hydroxy sandaracopimaric acid(1),16,17-dihydroxy-3-oxophyllocladane(2);2个酚苷:acte-oside(3),samioside(4);3个三萜:2α,3α,24-三羟基-乌索烷-12-烯-28-酸(5),2α,3α,19α-三羟基-乌索烷-12-烯-28-O-β-D-葡萄糖苷(6),2α,3α,19α,23-四羟基-乌索烷-12-烯-28-O-β-D-葡萄糖苷(7);4个黄酮:鼠李秦素(8),5-羟基-3,7,4′-三甲氧基黄酮(9),5-羟基-3,7,3′,4′-四甲氧基黄酮(10),木犀草苷(11)。与空白对照组相比,所有化合物均无明显缩短PT的作用(P<0.01),化合物3,4,7,10有显著缩短APTT的作用(P<0.01),化合物5对TT有明显的延长作用(P<0.01),化合物8对FIB有显著增加的作用(P<0.01)。结论:化合物2,4,10为首次从紫珠属植物中分离得到,化合物1,3,5,6,7,9为从裸花紫珠中首次分得。裸花紫珠可能是通过影响内源性凝血途径发挥止血作用。Objective: To study the hemostatic effect of chemical constituents from Callicarpa nudiflora.Method: The chemical constituents were isolated and purified via silica gel and Sephadex LH-20 column chromatography.Their structures were determined on the basis of spectral analysis.prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT)and fibrinogen(FIB)of the constituents rabbit blood samples were tested with rabbit blood in vitro.Result: Eleven compounds were isolated and identified as two diterpenens:7α-hydroxy sandaracopimaric acid(1),16,17-dihydroxy-3-oxophyllocladane(2).Two phenoic glycosides:acteoside(3),samioside(4).Three triterpenes:2α,3α,24-trihydroxy-ursa-12-en-28-oic acid(5),2α,3α,19α-trihydroxy-ursa-12-en-28-oic acid-28-O-β-D-glucopyranosyl ester(6),and 2α,3α,19α,23-tetrahydroxy-ursa-12-en-28-oic acid-28-O-β-D-glucopyranosyl ester(7).Four flavones: rhamnazin(8),5-Hydroxy-3,7,4′-trimethoxy-flavone(9),5-Hydroxy-3,7,3′,4′-tetramethoxyflavone(10),and luteoloside(11).All Compounds can not significantly shorten the PT(P〈0.01),compounds 3,4,7,10 can remarkedly increase APTT(P〈0.01),compound 5 can prolong the TT(P〈0.01)obviously,and compound 8 can significantly increase the contents of FIB(P〈0.01).Conclusion: Compounds 2,4 and 10 were isolated from this genus for the first time,and compounds 1,3,5,6,7 and 9 had been isolated from this plant for the first time.The hemostatic effect of C.nudiflora may be related to the activation of the intrinsic blood coagulation system.
