摘 要:目的研究Cox7a2调控睾酮生成的影响及其机制。方法构建Cox7a2荧光表达载体,转染TM3小鼠睾丸Leydig细胞。ELISA法测定上清液中睾酮含量,Western blot检测类固醇合成快速调节蛋白StAR表达,荧光分光光度计检测ROS水平。结果 Cox7a2抑制LH诱导的睾酮合成,降低StAR蛋白的表达,升高ROS水平。结论 Cox7a2通过抑制类固醇合成快速调节蛋白StAR的表达调控睾酮分泌,至少和增高ROS水平有关。Objective To investigate the regulation mechanism of steriodogenesis in Leydig cells by Cox7a2. Methods The recombinant expression vector pDsRed-Expression-N1-Cox7a2 was constructed and transfected into Leydig cells for Cox7a2 overexpression Level of testosterone in supernatant culture medium was measured by ELISA. The expression of steriodogenic acute regulatory protein (STAR) and reactive oxygen species (ROS) were detected by western blot and fluorometry. Results The recombinant expression vector pDsRed-Expression-N1-Cox7a2 was construct and transfected into Leydig cells suceesfully, Cox7a2 inhibited testosterone secretion of Leydig cells induced by LH treatment, and inhibited the expression of StAR protein. At the same time, Cox7a2 increased the activity of ROS in Leydig cells. Conclusion Cox7a2 inhibited testosterone production of Leydig cells patially due to ROS activity increase and down-regulation of StAR protein.
