摘 要:目的观察腹腔注射环磷酰胺对小鼠精子胞内钙离子浓度的影响。方法将20只雄性昆明小鼠随机分为对照组和模型组,按60mg/kg体重给对照组小鼠腹腔注射生理盐水(NS),另按60mg/kg体重给模型组小鼠腹腔注射环磷酰胺,每天1次,连续5d,常规饲养观察34d,然后对小鼠精子进行精液常规分析,对精子行FITC-AV/PI染色后用流式细胞仪检测精子凋亡率,用Ca^(2+)荧光探针Fluo-3/AM处理精子后用流式细胞仪检测精子胞内Ca^(2+)浓度。结果对照组与模型组精子密度分别为(5.20±1.34)×10~6/ml和(1.73±0.03)×10~6/ml,精子活力分别为(14.49±0.30)%和(6.64±1.88)%,精子活率分别为(68.39±1 5.1 3)%和(39.96±4.89)%,精子凋亡率(AV/PI)分别为(8.38±0.45)和(14.84±1.22),胞内Ca^(2+)浓度分别为(133.14±11.86)和(80.84±6.92),以上项目两组比较差异均有统计学意义(P<0.05)。结论腹腔注射环磷酰胺可使小鼠精子密度、活力、活率降低,精子凋亡增加,精子胞内Ca^(2+)浓度降低。造成精子胞内Ca^(2+)浓度降低的可能机理是环磷酰胺降低了精子特异性Ca^(2+)通道蛋白功能。Objective To investigate the change in concentration of calcium ion in mouse spermatozoa after peritoneal injection with cyclophosphamide. Methods 20 KUNMING male mice were randomly divided into the control group and the model group. During the first 5 days, mice in the control group received a peritoneal injection of NS with 60mg/kg weight once a day, while mice in the model group received a injection of cyclophosphamide, and then all the treated mice were normally breed for 34 days. The sperm count, motility and survival rate were measured, and the apoptosis of sperm stained with FITC-AV/PI was measured by flow cytometer, the concentration of calcium ion of sperm stained with Fluo-3/AM was also measured by flow cytometer. Results The sperm count, motility, survival rate, apoptosis rate and concentration of calcium ion of the control group were (5.20±1.34) × 10^6/ml, (14.49±0.30)%, (68.39± 15.13)%, (8.38±.45)and (133.14±11.86)respectively, whereas the above parameters of the model group were (1.73±0.03)X 106/ml, (6.64±1.88)%, (39.96±4.89)%, (14.84±1.22)and (80.84±.92)(P〈0.05). Conclusion After peritoneal injection with cyclophosphamide, the sperm count, motility, survival rate and the concentration of calcium ion were decreased, the apoptosis of sperm were increased.
