摘 要:[目的]内生真菌代谢产物球毛壳菌素A具有优异、广谱的抑菌活性,具备良好的新型杀菌剂的开发前景。为了更好的分析发酵过程地液中球毛壳菌素A的含量,采用反相高效液棚色谱法,建立了内生真菌代谢产物球毛壳菌素A定量分析方法。[结果]经梯度洗脱优化出的色谱条件:使用DuPont ZORBAX ODS C18色谱柱,以乙腈为流动相A、水为流动相B,梯度条件:(A)0~10min,20%-25%;10~20min,25%-40%;20—55min,40%~55%;55-75min,55%~75%;75~85min,75%~90%。流速为1.0mL/min,柱温:25℃,波长为254nm。[结论]该方法精密度为1.49%,重现性为5.41%,可以作为球毛壳菌发酵优化过程中代谢产物球毛菌素A的检测技术。[Aims] Chaetoglobosin A have good broad-spectrum bactericidal activity, which show good development prospects for the new fungicides. Here, the quantitative analysis method of chaetoglobosin A derived from endophytic fungus was developed using reversed-phase high performance liquid chromatography(RP-HPLC). [Results] The chromatographic conditions were optimized and obtained as follows: the separation was performed on a DuPont ZORBAX ODS C18 column. Acetonitrile(phase A) and ultrapure water(phase B) were used as mobile phase for gradient elution at a flow of 1.0 mL/min. The linear gradient of A was at 20-25%(0-10 min), 25-40%(10- 20 min), 40-55%(20-55 min), 55-75%(55-75 min), 75-90%(75-85 min), respectively. The column temperature was kept at 25℃ and the detection wavelength was 254 nm. [Conclusions] The method showed that the standard derivation was 1.49% and repeatability was 5.41%. Using the chromatographic conditions, further optimization of the metabolites of chaetomium fermentation process of detection was investigated.
