摘 要:丙型肝炎病毒(HCV)的基因分型在临床和流行病学的研究方面有重要意义。以多聚酶链反应的方法扩增HCV的5’-非编码区第28至284区段(HCV-J株),并以复合限制性内切酶水解的方法按Simmonds基因分型的标准,将HCV分为6型和若干亚型。结果:检测标本38份,均为1b型,与以往的检测结果类似。A study was described for identifying hepatitis C virus genotypes by restriction endonuclease cleavage of sequence amplified by polymerase chain reaction (PCR) from 5'-non-coding region,that the nucleotide sequence was from region 28 to 284 (HCV-J), GenBank, D10750. Using the enzymes of both Hae Ⅲ/Rsa I and of both Hinf I /Mva I, followed by cleavage with BstUI or ScrFI, according to the showed variant genotypes above. It was possible to identify and distinguish HCV genotypes as la, 1b, 2a, 2b, 3a, 3b, 4a and 6a. The method was used to investigate the prevalence of those genotypes in 38 cases of HCV infection in our hospital. All of the samples showed that they were genotype 1b, which was the same as what we have reported before in 1994. The method and its key innuence factors were also discussed.
