摘 要:目的:探讨四逆散对实验性溃疡性结肠炎(UC)大鼠白介素-1β(IL-1β)的影响,以及君臣配伍在组方中的作用。方法:将实验动物分为5组,采用免疫法造模,以大鼠结肠组织大体情况、结肠组织病理变化和IL-1β水平作为观测指标,用ELISA法检测大鼠结肠黏膜组织和血清IL-1β水平。结果:模型组大鼠结肠组织黏膜层可见溃疡形成,且有明显的炎症存在,认为本实验造模成功;模型组大鼠血清和结肠黏膜组织的IL-1β分别为(841.93±95.64)ng·L-1和(75.06±2.15)ng·L-1均显著高于正常组(P<0.01),灌服柴芍枳(1.31 g·kg-1)和柴芍(0.88 g·kg-1)后大鼠血清和结肠黏膜组织IL-1β显著降低(P<0.05),而灌服四逆散(1.75 g·kg-1)后大鼠结肠黏膜组织IL-1β(72.62±2.67)ng·L-1明显减少(P<0.01),但血清IL-1β(1015.52±176.47)ng·L-1却显著升高。结论:四逆散能够通过调节IL-1β来干预实验性UC,其全方发挥作用环节可能为抑制IL-1β由外周血单核细胞向炎症肠黏膜的移行;而方中君、臣药物配伍的作用环节为抑制IL-1β的分泌。Objective: To observe the influence of Sini Powders on interleukin-1 beta(IL-1β) of rats with experimental ulcerative colitis (UC) and effects of so-called the monarch drug and ministerial drug in the prescription. Method: The experimental animals were divided into 5 groups, and modeling was made by immunization. IL-1β levels in rats' colon mueosal tissues and serum were detected by ELISA. Result: Colon pathological changes with obvious inflammation were found after modeling. The levels of IL-1β in rats of the model group were higher than those in the normal group(P 〈 0.01 ). After feeding Chaishaozhi and Chaishao, IL-1β levels of UC rats were obviously reduced (P 〈 0. 05). After feeding Sini Powders, IL-1β levels in UC rats' colon mueosal tissue were markedly reduced (72.62± 2. 67)ng· L^-1, (P 〈 0. 01 ), but they were obviously increased in serum (1015. 52± 176. 47)ng·L^-1. Conclusion: We speculate that Sini Powders can interfere with experimental UC by adjusting IL-1β levels. Sini Powders may play a role in inhibiting moving of IL-1β from the mononuclear cells in peripheral blood to intestinal mucosa, and the compatibility of the monarch drug and ministerial drug play a role in suppressing secretion of IL-1β.
