摘  要:目的评价鼠疫菌检测技术在鼠疫实际监测工作的应用效果。方法分别用细菌培养、反向血凝、金标、PCR和ELISA方法检测染疫动物脏器材料F1抗原,比较了不同方法在实际监测工作中的差异。结果细菌培养、ELISA、R IHA、PCR、金标阳性检出率分别为4.68(、8.19(、15.79(、15.20(、14.62(;以标准值为对照,敏感度和特异度分别为细菌培养:敏感度36.4%、特异度100%;ELISA敏感度63.6%、特异度100%;R IHA敏感度72.7%、特异度92.6%;PCR敏感度95.5%、特异度96.6%;金标敏感度81.8%、特异度95.3%。结论细菌培养在实际应用中,受到材料腐败程度影响较大,但能培养出鼠疫菌。反向血凝在应用中以其方便、迅速且成本较低,在大范围的疫源监测中起到重要作用。胶体金法在应用中简单、准确、便捷,适用于现场操作。PCR技术在非典型鼠疫菌的检测中意义重大[1]。ELISA技术方便、快捷、特异性强,但对材料要求较高。Objective To evaluate the application effect of examination technology in actual detection of Yersinia pestis.Methods Organs of infected animals by bacterial culture,Using bacterial culture RIHA,gold colloid,PCR and ELISA to detect the F1 antigen in organ materials of infected animals,and compare the differences of these methods in actual detection.Results The positive rate of Bacterial culture,ELISA,PCR,RIHA and gold standard were 4.68%,8.19%,15.79%,15.20% and 14.62% respectively.The standard values as control,the sensitivity and specificity of these methods were as follow,Bacterial culture: 36.4%,100%,ELISA: 63.6%,100%,RIHA: 72.7%,92.6%,95.5%,96.6%,and gold colloid: 81.8%,95.3%.Conclusions In actual detection,using bacterial culture could culture pure Y.pestis although it was influenced by the material level of corruption badly.RHIA is convenient,fast and low cost,and plays an important role in a wide range of foci monitoring.gold colloid is simple,accurate,convenient,and is suitable for field operations.PCR is significant in the detection of atypical Y.pestis.ELISA is convenient,fast,and high specificity,but requires better materials.
