摘 要:在环境释放条件下,研究了3种转基因青蒿品系及其亲本品种的农艺性状,分析了各个农艺性状指标之间的相关性。利用实时荧光定量逆转录PCR技术检测3种转基因青蒿目的基因表达变化差异,并利用HPLC-ELSD技术测定青蒿材料中的青蒿素含量。研究表明:在田间栽培条件下,转基因青蒿植物个体发育正常;转基因青蒿在株高、冠幅、茎粗、叶间距、叶形、叶片宽度方面与其受体材料无显著差异,但在个别时间点存在例外;转基因青蒿和其亲本材料的生长趋势大致相似;3种转基因青蒿目的基因表达变化显著,转基因青蒿中青蒿素含量明显增高,与预期结果一致。The agronomic traits of 3 transgenic Artemisia annua lines and their parents were studied under environmental release conditions,and the correlations between their agronomic traits were analyzed.The variation of the target gene expression in the 3 transgenic lines was estimated by realtime fluorescent quantitation RT-PCR,and the artemisinin contents of the 3 transgenic lines were determined by HPLC-ELSD(HPLC-evaporative light-scattering detection).The results showed that in field trials the transgenic plants grew and developed normally and were not significantly different from control plants in plant height,crown width,stem diameter,leaf spacing,leaf shape and leaf width except some points of time.The growth rates of both transgenic and control plants were also approximately similar.The 3 transgenic lines were significant in variation of target gene expression and obviously increased in artemisinin content,being in line with the anticipated result.
