摘  要:目的为探讨X线照射新生大鼠后对小脑皮质浦氏细胞层排列的影响。方法将新生一日(P1)大鼠经X线全身照射(剂量为1.5Gy),照射后3周取小脑用于Nissl染色及免疫组化染色分析。Calbindin-D28k染色和GFAP染色标记浦氏细胞及在小脑发生过程中起重要作用的贝格曼神经胶质细胞。结果 X线照射的新生大鼠3周后可见小脑变小,部分小脑叶排列紊乱。与对照组相比,Calbindin D-28k阳性细胞未能形成单层排列,部分细胞侵入颗粒细胞层,且树突发育不良。同时可见GFAP阳性细胞纤维未能通过分子层而达软膜下。结论 X线照射新生大鼠后,贝格曼神经胶质细胞纤维的排列紊乱导致小脑皮质浦氏细胞层排列的异常。Objective To investigate the abnormal align of Purkinje cells in rats following neonatal Xirradiation. Methods Wistar rats were exposed to X-irradiation (1.5 Gy) on postnatal day 1. At 3 weeks of age, the cerebella were examined by Nissl staning and immunohistochemical staining. The histological changes in Purkinje cells and Bergmann glial fibers which play an important role in the development of cerebellum were observed by calbindin-D28k (CB) and glial fibrillary acidic protein (GFAP) immunohistochemistry. Results The cerebella of rats exposed to X-irradiation showed smaller and abnormal foliations compared with controls. In X-irradiatied rats, CB-immunoreactive (IR) Purkinje cells did not exhibit monolayer align, some cell bodies invaded into the granular layer or white matter, and the dendrites were disoriented. On the other hand, GFAP-IR Bergmann glial fibers had not extended their processes into the molecular layer perpendicular to the pial surface compared with controls. Conclusion The disorder of Bergmann glial fibers results in the abnormal align of Purkinje cells in rats following neonatal X-irradiation.
