摘 要:利用扩增片段长度多态性(Amplified fragment length polymorphism,AFLP)标记技术对桃[Prunus persica (L.) Batsch]红肉材料及其F1代共41份种质材料进行了DNA多态性分析,从64对(E+2/M+3)引物组合中筛选出7对引物用于扩增基因组DNA,共获得清晰可辨的325个标记,其中多态性标记为113个,多态性检出率为34.77%。根据AFLP扩增结果,利用NTSYSpc2.10e软件DICE进行相似系数分析,绝大多数材料的遗传相似系数集中在0.80~0.99,说明各种质材料之间遗传多样性不太丰富。通过UPGMA进行聚类分析,在遗传相似系数约0.88处,有9份红肉桃种质材料可聚为一大类,24份杂交一代与其母本金华大白桃和杭蜜2号可聚为第二大类。040222、红肉桃1、粘核红肉桃、红肉桃10、红肉桃18与040305这6份材料分别单独聚为一类,且与其他种质材料的遗传相似系数相对较小,表明其亲缘关系较远。41 varieties of red-flesh peach [Prunus persica(L.) Batsch] and their F1 hybrid were analyzed by AFLP markers. Seven pairs of primers were selected out of 64 pairs of primers(E+2/M+3) to amplify the genomic DNA. Totally 325 AFLP bands were obtained, 113(34.77%) of them were polymorphic markers. The DICE similarity coefficient of the AFLP amplification results were analysed. The DICE similarity coefficient of the AFLP amplification results were analyzed by NTSYSpc2.10e software,and the genetic similarity coefficient was 0.80~0.99, suggesting a deficient genetic diversity. The clustering dendorgram was constructed by UPGMA. When the similarity coefficient was 0.88,9 red-flesh peach materials were clustered into one group; and 24 F1 hybrids and their female parents were clustered into another group. 040222, Hongroutao No.1, Nianhehongroutao, Hongroutao No.10, Hongroutao No.18 and 040305 were clustered into six different groups, and had relatively small genetic similarity coefficient, indicated a further relatioship.
