摘 要:研究了不同冬性大白菜[Brassica campestris L. ssp. pekinensis(Lour.)Olsson]萌动种子在2℃低温处理过程中,其硝酸还原酶(Nitric acid reductase,NR)活性、可溶性蛋白质含量及过氧化物酶(Peroxide enzyme,POD)同工酶、细胞色素氧化酶(Cytochrome oxidase,COD)同工酶和β-酯酶(Beta-esterase isozyme,β-EST)同工酶的电泳图谱变化。结果表明,萌动种子NR活性与大白菜的耐抽薹性密切相关,即随着耐抽薹性减弱NR活性降低,极耐抽薹品种萌动种子的NR活性是易抽薹品种的3~4倍,萌动种子中NR活性的高低在一定程度上可反映大白菜的耐抽薹特性。在春化过程中,极耐抽薹品种的萌动种子NR活性表现为先快速下降,然后维持一段低活性水平;易抽薹品种萌动种子的NR活性表现与之相反,先是快速升高,到春化处理的15d达最大值,然后快速下降。不同大白菜品种萌动种子可溶性蛋白质含量的变化趋势基本一致,表现为"上升-下降-上升"的倒"S"型变化;大白菜萌动种子在完成春化的时候陆续有Rf为0.788的特异蛋白出现。不同大白菜品种萌动种子的POD、COD和β-EST同工酶酶谱基本相同;春化20d以后,所有品种Rf为0.632的β-EST同工酶带从大白菜中消失,而POD和COD同工酶活性有所增加,且各材料均陆续出现自己特有的POD和COD同工酶带。The NR (Nitric acid reductase)’s activity, content of soluble protein, the changes of PAGE of the isozyme of POD (Peroxide enzyme), COD(Cytochrome oxidase) and β-EST(Betaesterase isozyme) in germinating seeds of Brassica campestris L. ssp. pekinensis(Lour.) Olsson were detected during vernalization at 2℃. The results showed that the NR’s activity in germinating seeds was closely related to bolting tolerance of B. campestris ssp. pekinensis. The activity of NR in germinating seeds of varieties very resistant to bolting is 3~4 times of varieties’ easy bolting. The level of NR’s activity can reflect the characteristics of bolting tolerance of Brassica campetris L. ssp. pekinensis(Lour.) olsson to some extent. During the low temperature treatment, the NR’s activity dropped first, then kept at a low activity level in the varieties very resistant to bolting; but the NR’s activity increased rapidly first, and reached the highest activity at 15 days, then decreased quickly in the varieties easy bolting; The change tendency of soluble protein content were consistent among different varieties of B. campestris ssp. pekinensis, which presented as inverse "S" curve of "rise-down-rise", and a new protein band with Rf=0.788 were detected at the end of vernalization of each cultivar; The zymograms of POD, COD and β-EST were consistent among different B. campestris ssp. pekinensis also, since twenty days of vernalization, the activity of POD and COD increased, and special bands of POD and COD isozyme in some varieties were appeared, but the common β-EST isozymes of Rf=0.632 in all cultivals were disappeared.
