雷公藤胃漂浮缓释微丸的制备

摘　　要：目的:制备雷公藤胃漂浮缓释微丸,考察其漂浮和释药性能。方法:以微丸的体外释放度、漂浮性能为考察指标,采用挤出滚圆法制备胃漂浮空白微丸,采用流化床包衣设备依次进行载药层和缓释层包衣,制备雷公藤胃漂浮缓释微丸。结果:以90%十八醇为助漂剂,10%微晶纤维素为稀释剂制得空白胃漂浮微丸;以1%羟丙基甲基纤维素和0.5%十二烷基硫酸钠为载药层包衣材料,以含3%聚维酮的乙基纤维素水分散体为缓释层包衣材料。制得的雷公藤胃漂浮缓释微丸在人工胃液中立即起漂,8h漂浮率均>80%,体外释药机制符合一级释药方程。结论:所制雷公藤胃漂浮缓释微丸具有良好的漂浮性能和缓释特性。OBJECTIVE： To prepare gastric floating sustained-release pellets of Tripterygium wilfordii,and to study their behavior of floating and release characteristics. METHODS： The gastric floating blank pellets were prepared by extrusion-spheronization method with release rate and floating characteristics as factors. The drug-loading layer and sustained-released layer of pellets were coated using fluid-bed coater to prepare gastric floating sustained-release pellets of T. wilfordii. RESULTS： The gastric floating blank pellets were prepared with 10% MCC as diluent and 90% cetylalcohol as floating assistant. 1% HPMC and 0.5% SDS were used as drug- loading layer and ethylcellulose aqueous dispersion containing 3% PVPK30 as sustained-released layer. The gastric floating sustained-release pellets of T. wilfordii. could float immediately in simulated gastric fluid with 8 h floating rate more than 80%. The release behavior of pellets was fitted to first-order equation. CONCLUSION： The prepared pellets have sound floating behavior and sustained release characteristics.
