丹蒲胶囊对自身免疫性前列腺炎模型IL-2、IL-8、IL-10及NF-κB的影响

摘　　要：目的:探讨丹蒲胶囊治疗慢性前列腺炎的分子免疫机制。方法:90只Wistar大鼠随机分为丹蒲胶囊高、中、低剂量组、前列泰组、模型组和正常组6组。采用纯化大鼠前列腺蛋白结合免疫佐剂法建立实验性自身免疫性前列腺炎大鼠模型,连续给药10周。ELISA法测定大鼠前列腺组织及血清细胞因子IL-2、IL-8、IL-10及NF-κB变化。结果:模型组前列腺组织及血清IL-2、IL-8、IL-10水平明显高于正常组(P<0.05),各中药干预组前列腺组织及血清IL-2、IL-8、IL-10水平均低于模型组(P<0.05),以丹蒲胶囊高剂量组为优(P<0.01)。模型组前列腺组织NF-κB水平明显高于正常组(P<0.05),丹蒲胶囊各剂量组前列腺组织NF-κB水平明显降低,以高剂量组效果最佳(P<0.05)。结论:丹蒲胶囊通过对IL-2、IL-8、IL-10、NF-κB的调节而发挥治疗作用。Objective To explore the action of Dan Pu（丹蒲） capsule in the treatment of chronic prostatitis. Methods Ninety Wistar male rats were divided into 6 groups randomly： Dan Pu） capsule group with high dosage, Dan Pu capsule group with middle dosage, Dan Pu Capsule group with low dosage, Qian Lie Tai（前列泰） group, the model control group and normal group. The experimental autoimmune prostatitis （EAP） animal model was established by the method of modified prostate protein with immunologic adjuvant. The levelof interleukin 2 （IL-2）, interleukin8 （IL-8）, interleukin 10 （IL-10） and nuclear factor kappa B （NF-κB） were measured by enzyme linked immunosorbent assay （ELISA）. Results The level of IL-2, IL-8 and IL-10 in the model group was higher than that of the normal group （P〈0.05） while, it was obviously decreased in the treatment group with TCM （P〈0.05）, and the reduced degree in the Dan Pu group with high dosage were most significant. Meanwhile, the level of NF-κB in the model group was higher than that of the normal group （P〈0.05）. It was obviously declined in the treatment group with TCM, and that of the Dan pu group with high dosage was the best （P〈0.05）. Conclusion Dan Pu capsule has the action of lowering the level of IL-2, IL-8,IL-10 and NF-κB, which can be one of the mechanisms on Dan Pu capsule in the therapy of Chronic prostatitis.
