摘  要:采用正交实验法对黄连中小檗碱盐析提取的浸泡时间、黄连粒度、硫酸浓度、HCl调pH值、石灰乳调pH值及NaCl用量等影响因素进行了优化。确定最优的提取方案为:浸泡时间24 h、黄连粒度0.355~0.5 mm、硫酸浓度0.40%,固液比1∶16、石灰乳调pH值12、HCl调pH值5、NaCl用量25%,在此条件下,小檗碱产率为8.4%、纯度为72.7%。各因素对黄连小檗碱提取效果影响大小依次为:固液比>硫酸浓度>浸泡时间>黄连粒度>NaCl用量>石灰乳调pH值>HCl调pH值。Optimization of salting-out extraction of berberine from Huanglian by orthogonal experiment was carried out.The effects of immersion time,Huanglian particle size,the concentration of sulfuric acid,solid-liquid ratio,the pH value adjusted by lime milk,the pH value adjusted by HCl,NaCl amount on the extraction were investigated.The optimum extraction conditions were as follows: immersion time 24 h,Huanglian particle size 0.355~0.5 mm,the concentration of sulfuric acid 0.40%,solid-liquid ratio 1∶16,the pH value adjusted by lime milk 12,the pH value adjusted by HCl 5,NaCl amount 25%.Under above conditions,berberine with yield of 8.4% and purity of 72.7% was obtained.The effect of various factors on berberine extraction was in the order of solid-liquid ratiothe concentration of sulfuric acid 〉immersion time〉Huanglian particle size〉NaCl amount〉the pH value adjusted by lime milk〉the pH value adjusted by HCl.
