摘  要:目的:为建立一种稳定的大鼠胎鼠海马神经元培养方法并能够有效的鉴定其纯度。方法:分离Wista大鼠胚胎并取海马组织,比较两种常用酶,胰酶和木瓜酶消化效果对海马神经元产量的影响及两种不同培养方法对海马神经元纯度的影响。结果:培养的神经元胞体清晰晕光明显,应用两种不同的酶消化对神经元的产量影响无明显统计学意义,而应用无血清方法培养神经元在纯度上要优于应用阿糖胞苷去除神经胶质细胞的培养方法。结论:采用木瓜酶消化并用无血清培养的方法培养大鼠胎鼠海马神经元,稳定可靠,提高了原代神经元培养的产量和纯度。Objective:To devel op a suitable primary culturing method of c ortical neurons of newborn Wista rats and to es tablish an experiment cell model of the neurons.Method:The hippocampus of newbo rn Wista rats were dissected and digest ed with two kinds of enzymes to investig at the quantity and purity of the cult ured neurons.Results:The neurons cultu red by the two kinds enzyme were all cl ear and health,they were not different significantly.However,the purity of ne urons cultured with neurobasal was high er than neurons cultured with arabino si de cytarabine(Ara-C).Conclusion:The me thods dissected hippocampus neurons dig ested with papain enzyme and cultured wi th neurobasal are stab ilization and safety so increase the qua ntity and purity of neurons.
