GOEXISTENCE OF IMMUNE NEURO ENDOCRINE SUBSTANCES IN THE RAT CENTRAL NEURONS

摘  要:目的研究白细胞介素-2、代谢型谷氨酸受体1亚型(mGluR1)和雌激素受体(ER)在中枢神经系统神经细胞的表达,证实它们共存的可能性。方法使用种属特异性1抗(山羊抗IL-2、兔抗mGluR1、小鼠抗ER血清)的免疫细胞化学三重标记技术,在相邻的两张连续切片(1张显示IL-2,另1张显示mGluR1和ER)上证实大鼠大脑皮质、延髓和脊髓内上述3种物质的共存。结果在上述部位的中枢神经元内可见IL-2、mGluR1和ER3种免疫反应性标记细胞。IL\|2免疫反应产物为棕褐色,位于胞浆内。相邻切片的mGluR1免疫反应产物为蓝黑色,位于胞膜;ER免疫反应产物亦为棕褐色,位于胞浆或核内。显示mGluR1和ER的同一切片上有mGluR1/ER双标细胞,双标细胞约占全部标记细胞的50%~60%。通过对相邻的两张切片的投影核对,证实存在mGluR1/ER/IL-2三标细胞,三标细胞占mGluR1/ER双标细胞的30%。结论在大鼠中枢神经系统内mGluR1、ER和IL-2可共存于同一个神经细胞,从而首次在细胞水平为神经免疫内分泌网络学说提供了形态学依据。Objective\ To investigate the expression of Interleukin 2(IL 2),metabotropic glutamate receptor subunit 1(mGluR1) and estrogen receptor (ER) in neurons of the rat central nervous system (CNS) and identify the coexistence possibility of these immune neuro endocrine substances in the central neurons.Methods\ The triple labeling immunocytochemical staining technique with different species specific primary antibodies(goat anti IL 2 serum,rabbit anti mGluR1 serum and mouse anti ER serum)were used to incubate two serial neighbor sections(one for demonstrating IL 2,another for mGluR1 and ER) and identify the coexistence of above three kinds of substance in the cerebral cortex,medulla oblongata and spinal cord.Results\ There were IL 2, mGluR1 |and ER immunoreactive(ir)labeled neurons in the above mentioned central areas.The IL 2 ir production showed brown in color,locating in the cytoplasm.In the neighbor serial section,mGluR1 ir production showed blue black in color,locating at the cell membrane;the ER ir production also showed brown color,locating in the cytoplasm or nucleus.There were mGluR1/ER double labeled cells in same section, which accounted for about 50% 60% of the total single and double labeled neurons.It was identified by projection check of serial neighbor sections that there were mGluR1/ER/IL 2 triple labeled cells which accounted for about 30% of total mGluR1/ER double labeled nuurons.Conclusion\ mGluR1,ER and IL 2 can coexist in same rat central neurons,therefore,providing morphological basis for the theory about immune neuro endocrine network at the cellular level for the first time.
