PTEN和p38MAPK在前列腺良恶性病变中的表达及意义

摘　　要：目的:探讨PTEN蛋白及丝裂素活化蛋白激酶p38(p38MAPK,p38)在前列腺良恶性病变组织中的表达差异及其临床意义。方法:应用免疫组织化学SABC法检测20例良性前列腺增生(BPH)组织、20例前列腺上皮内瘤(PIN)组织及46例前列腺癌(PC)组织中的PTEN和p38的表达。结果:PTEN在BPH、PIN和PC组织中表达活性递减,差异有统计学意义(P<0.01);p38在BPH、PIN和PC组织中表达活性递增,差异有统计学意义(P<0.01);PTEN的表达与前列腺癌患者临床分期、病理分级呈负相关(r分别为-0.352和-0.653,P<0.05),而p38的表达则与上述项目呈正相关(r分别为0.298和0.596,P<0.05);PTEN与p38表达呈负相关(r=-0.602,P<0.01)。结论:PTEN蛋白低表达和p38活性的增强可能参与了前列腺组织的恶化,在前列腺肿瘤的发生及发展中起重要作用。Objective：To study the different expression and clinical significance of PTEN and p38 mitogen-activated protein kinase（MAPK） in benign and malignant prostatic lesions.Methods：SABC immunohistochemical analysis was used to determine the expression levels of PTEN and p38MAPK in 20 cases of benign prostatic hyperplasia（BPH）,20 cases of prostatic intraepithelial neoplasia（PIN） and 46 cases of prostatic carcinoma（PC）.Results：The expression of PTEN was the highest in BPH group and significantly higher in PIN group compared with that of PC group（P 0.01）.The expression of p38MAPK was the highest in PC group and significantly higher in PIN group compared with that of BPH（P 0.05）.The expression of PTEN protein in prostatic carcinoma was negatively correlated with tumor stage and histological grade（r =-0.352,-0.653,P 0.05）;While the expression of p38MAPK was positively correlated with the above features（r = 0.298,0.596,P 0.05）.There was a negative correlation between expressions of PTEN and p38MAPK（r =-0.602,P 0.01）.Conclusion：The lower expression of PTEN protein and elevated activation of p38MAPK are involved in the malignant transformation of prostatic epithelium and may play a great role in the genesis and development of prostatic tumor.
