摘  要:目的:比较分析结核性胸膜炎患者血清与胸腔积液蛋白质组的异同,为探讨结核性胸膜炎的发生机制和寻找诊断标志物提供实验依据。方法:采用弱阳离子交换蛋白质芯片(WCX2)处理分析样本、应用表面增强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术,以蛋白质/多肽质荷比(m/z)区分不同的蛋白质/多肽,对5例结核性胸膜炎患者血清与胸腔积液蛋白质组进行描述性分析。结果:(1)在5例患者血清中检测到134种血清蛋白质/多肽,其中76种在胸腔积液中未出现;在5例患者胸腔积液中检测到269种蛋白质/多肽,其中211种在血液中未出现。(2)在5例患者血液中均检测到m/z分别为2660、3191、4208、5903、7764、9287等6种蛋白质/多肽;在5例患者胸腔积液中均检测到m/z分别为2416、2660、2900、2940等4种蛋白质/多肽;在5例患者血液和胸腔积液中均检测到m/z为2660的蛋白质/多肽。结论:结核性胸膜炎患者胸腔积液蛋白质组远比其血清蛋白质组复杂,对其进行深入研究有助于明晰结核性胸膜炎的发生机制和寻找诊断标志物。Objective To compare the difference of proteome between in serum and in pleural effusion in patients with tuberculous pleurisy,so as to provide experiment evidences for studying pathogenetic mechanism and finding diagnostic markers of tuberculous pleurisy. Methods The proteome of serum and pleural effusion in 5 patients with tuberculous pleurisy were detected by weak cation exchange protein chip (WCX2) and surface-enhanced laser desorption-ionization time of flight mass spectrometry(SELDI-TOF-MS). And different proteins/polypetides were analyzed descriptively according to the difference of m/z. Results There were 134 kinds of proteins/polypeptides in serum and 76 kinds of them did not exist in pleural effusion in 5 patients with tuberculous pleurisy,while 269 kinds of proteins/polypetides were found in pleural effusion and 211 kinds of them did not exit in serum. Six kinds of proteins/polypetides whose m/z were 2 660,3 191,4 208,5 903,7 764,and 9 287 respectively were found in serum in all patients,while 4 kinds of proteins/polypetides whose m/z were 2 416,2 660,2 990,and 2 940 respectively were found in pleural effusion in all patients. The protein/polypetide whose m/z was 2 660 was found in both serum and pleural effusion in all the 5 patients. Conclusion The proteome in pleural effusion is more complicated than that in serum in patients with tuberculous pleurisy,on which more progressive research will benefit to understanding the pathogenetic mechanism and finding the diagnostic markers of tuberculous pleurisy.
