摘 要:目的体外研究大鼠脊髓损伤后表达Nogo蛋白受体(Nogo receptor,NgR)的巨噬细胞的吞噬作用及其对损伤神经元再生的作用。方法将髓鞘碱性蛋白(myelin basic protein,MBP)分别加入表达NgR的巨噬细胞、空载巨噬细胞及正常巨噬细胞中,应用Western blot比较这3组巨噬细胞的吞噬作用。将3组巨噬细胞加入损伤神经元中培养,采用MTT法及测定乳酸脱氢酶(lactate dehydrogenase,LDH)的含量,比较3组巨噬细胞对损伤神经元轴突再生的作用。结果 Western blot结果显示表达NgR的巨噬细胞组吞噬作用与其他2组比较差异有统计学意义(P<0.05)。MTT比色法及LDH含量测定结果显示表达NgR的巨噬细胞组吞噬作用与其他2组比较差异有统计学意义(P<0.05)。结论 NgR的表达在体外可增强巨噬细胞的吞噬作用,表达NgR的巨噬细胞可促进神经损伤后的轴突再生。Objective To study the phagocytic capacity and nerve regeneration on injured neurons of macrophages expressing Nogo receptor (NgR) in vitro after spinal cord injury in rats. Methods The myelin basic protein (MBP) was added into macrophages expressing NgR, no-load macrophages and normal macrophages. Western blot was used to compare the phagocytic capacity of macrophages of the 3 groups. Each of the 3 macrophages groups was cultured with the injured nerve. The micro-MTT colorimetry and lactate dehydrogenase (LDH) assay were used to detect the growth of axons to compare the effect of macrophages on axon regeneration in the 3 groups. Results Western blot results showed that there were differences between the group of macrophages NgR with the other 2 groups in phagocytic capacity, and the differences were statistically significant. (P 〈 0.05 ). The results of MTT assay and detection of LDH content showed that there were differences between the group of macrophages expressing NgR with the other 2 groups, and the differences were statistically significant. (P 〈 0.05). Conclusion Expression of NgR in macrophages can enhance the phagocytic capacity of macrophages. The macrophages expressing NgR can promote axon regeneration after nerve injury.
