5-脂氧化酶、白三烯在动脉粥样硬化大鼠主动脉中的表达

摘　　要：目的探讨5-脂氧化酶(5-LO)、白三烯B4(LTB4)、白三烯C4(LTC4)在动脉粥样硬化大鼠主动脉及血液中的表达及其意义。方法选取正常雄性SD大鼠25只,随机分为正常对照组和动脉粥样硬化组。正常对照组普通饲料喂养,动脉粥样硬化组高脂饲料喂养建立动脉粥样硬化大鼠模型,14周后处死大鼠,采血,全自动生化仪检测血脂,ELISA法检测血清及主动脉中5-脂氧化酶、白三烯B4、白三烯C4的含量。结果动脉粥样硬化组血脂较正常组明显升高(P<0.01),动脉粥样硬化组血液中及主动脉中5-脂氧化酶、白三烯B4、白三烯C4的含量较正常组升高(P<0.05)。结论 5-LO、LTB4、LTC4在动脉粥样硬化大鼠主动脉及血液中表达均增高,5-LO/LTS通路可能是动脉粥样硬化潜在的干预靶点。Objective To prove the abnormal expression of 5-LO,LTB4,LTC4 in aorta and blood of Atherosclerotic rats,discuss the meaning of 5-LO/LTs role in Atherosclerosis.Methods Twenty-five normal SD rats were divided into two groups randomly,one was normal control group（n=10）,the other was atherosclerotic group（n=15）.Normal control group was fed with normal diet,Atherosclerotic group group was fed with high cholesterol diet in order to found the model of atherosclerotic rats.fAter 14 weeks,blood lipids were measured,and 5-LO,LTB4,LTC4 in blood and aorta were measured too.Results The level of blood lipid in atherosclerotic group was higher than that in normal control group.The level of 5-LO,LTB4,LTC4 in blood and artor of atherosclerotic rats was higher than that in normal control group.Conclusions The expression of 5-LO,LTB4,LTC4 in blood and artor of atherosclerotic rats was abnormal,the 5-LO/LTs role was a potential target in treatment of atherosclerosis.
