HPV16(E6-E7)转基因小鼠肿瘤细胞系的建立与生物学特性观察

摘  要:对转基因小鼠HPV16(E6-E7)所形成的肿瘤进行原代培养,体外传代观察,传40代后对培养细胞的形态学、生长动力学、免疫组织化学、细胞核型和致瘤性等生物学特性进行分析。结果表明,该细胞系已在体外培养生存1年以上,传120代;细胞形态多样,以多角形、短梭形为主;染色体核型为非整倍体,众数58~62条;软琼脂集落形成率为18.5%;细胞体外生长倍增时间为47.2 h;免疫组织化学检测到HPV16-E6蛋白,而未检出p53;裸鼠移植成瘤率为100%。该研究成功建立了转基因小鼠HPV16(E6-E7)肿瘤细胞系,为HPV发病机制的研究和治疗药物筛选提供了新的可靠材料。The tumor tissue from HPV16(E6-E7) transgenic mice was used for cell culture in vitro.After 40 passages,the cell morphology,growth kinetic,Karyo-type analysis,immunohistochemical analysis and oncogenicity of the cell line were investigated,respectively.The results showed that this cell line had been passaged 120 times within 1 year.Cells were short spindled to polygon morphologically.Karyo-type analysis revealed that HPV16(E6-E7) cells were aneuploidy with a median chromosome number of 58-62.The cloning efficiency in soft agar averaged 18.5% and the population doubling time was 47.2 h.The HPV16-E6 protein expression and the absence of p53 were verified by immunohistochemical analysis.Tumor formation rate in nude mice was 100%.Furthermore the pathological analysis showed that the HPV16(E6-E7) cells were originated from fibrosarcoma.
