鼻息肉中Survivin基因表达及其与Ki67表达的相关性分析

摘　　要：目的研究Survivin在鼻息肉组织中表达及其与Ki67表达的相关性,探讨Survivin在鼻息肉发病过程中的作用。方法收集28例鼻息肉组织(鼻息肉组)和12例正常下鼻甲组织(对照组),用免疫组化法检测鼻息肉组与对照组组织中Survivin与Ki67的表达,观察比较两组之间的表达差异。结果免疫组织化学染色显示Survivin、Ki67免疫阳性细胞数及着色强度均明显高于对照组。图像分析显示,鼻息肉组Survivin、Ki67的积分光密度(IOD,103/HP)分别为50.21±6.32、18.25±2.71,与对照组5.67±0.58、3.80±0.37相比,有显著性差异(P<0.05);Spearman等级相关分析显示,鼻息肉中Survivin表达与Ki67表达密切相关(r=0.650,P<0.01)。结论鼻息肉组织中Survivin、Ki67表达均上调,且二者有协同表达关系,鼻息肉可能是通过凋亡抑制基因Survivin使炎性细胞凋亡受到抑制,从而促进炎性细胞浸润生长,导致一种慢性炎症反应,并造成局部组织增生。Objective To study Survivin and Ki67 expression in nasal polyps and their correlation,and to explore the role of Survivin in the pathogenesis of nasal polyps.Methods 28 cases of nasal polyps and 12 cases of normal inferior turbinate mucosa were enrolled in this study.Immunohistochemistry method for Survivin and Ki67 were performed in two groups.Results The numbers and intensity of positive stained cells of Survivin and Ki67 were higher in the nasal polyps than that in the control group.IOD of Survivin（10^3/HP）in the nasal polyps was 50.21±6.32、18.25±2.71compared with 5.67±0.58、3.80±0.37 of the control group by image analysis.There were significant differences between two groups（P〈0.05）.Spearman correlation test showed that there was a close correlation between the Survivin and Ki67 expression in nasal polyps（r=0.650,P〈0.01）.Conclusion The results showed that Survivin and Ki67 were over expressed in nasal polyps,and there was a close correlation between each other,Survivin might inhibit apoptosis of inflammatory cells including eosinophils to promote infiltration and growth of inflammatory cells,which lead to the chronic inflammatory reaction and hyperblastosis in nasal polyps.
