摘  要:研究活性氧(ROS)-过氧化氢和抗氧化剂-还原型谷胱甘肽(GSH)对小鼠2-细胞期胚胎体外发育的影响.用含有不同浓度H2O2的mSOF液培养小鼠胚胎,从H2O2浓度0.14 mg/L组开始的囊胚发育率显著低于对照组和其它组(P<0.05);以mSOF液为基础液,在添加0.14 mg/L H2O2的基础上分别添加不同浓度的GSH,阳性对照组的囊胚发育率显著高于阴性对照组(P<0.05);在添加外源性H2O2(0.14 mg/L)的情况下,随着GSH浓度的增加其囊胚发育率也显著增加.结果表明,H2O2可诱导小鼠2-细胞期胚胎体外发育的阻滞,GSH可阻止H2O2对小鼠2-细胞期胚胎的抑制效果.Studied the effect of ROS-hydrogen dioxide and antioxidant-GSH on the development of 2-cell embryo of mouse in vitro.Cultured the embryo of mouse in mSOF medium with different concentration H2O2,through the analysis on the blastula developing rate,control group and the groups of 0.12 mg/L,0.13 mg/L concentration were significantly higher than other groups(P 0.05);with mSOF as the basic medium,and then adding 0.14 mg/L H2O2 and different concentration GSH in,positive control group was significantly higher than negative control group(P 0.05);and on the situation of exogenous hydrogen dioxide(0.14 mg/L) was added in,with the increase of the concentration of GSH,the blastula developing rate also increased significantly.The results showed that the ROS(such as H2O2) can inhibit the development of embryos in vitro,GSH can prevent the development block of 2-cell embryo of mouse caused by H2O2.
