摘 要:用超临界流体快速溶胀技术和超临界流体辅助雾化技术耦合法制得胰岛素-三棕榈酸甘油酯复合微粒。考察了操作温度和胰岛素溶液流量对复合微粒的形貌和粒径的影响。结果表明,45℃下得到的复合微粒平均粒径为25μm,50℃下(不同胰岛素溶液流量)得到的复合微粒平均粒径为3.9μm。体外释放试验表明,50℃时得到的复合微粒具有较好的缓释效果,120 min时累积释放率为36%,释放完全约需112.5 h。The tripalmitin composite particles loaded with insulin were prepared by a process combining supercritical fluid-assisted atomization with rapid expansion of supercritical solution.The effects of the operating temperature and flow rate of insulin solution on the appearance and diameter of the products were investigated.The results showed that the composite particles obtained at 45℃had an average size of 25μm.The particles prepared at 50℃under different flow rates of insulin solution had an average size of 3.9μm.The in vitro test showed that the products prepared at 50℃had an obvious sustained release behavior.The accumulative release at 120 min was 36%and the insulin was completely released within 112.5 h.
