摘  要:目的:研究鱼腥草抗病毒有效部位槲皮素体外抗人巨细胞病毒(HCMV)的作用及其作用机制。方法:用鱼腥草抗病毒有效部位槲皮素配成不同浓度的溶液,噻唑蓝比色法(MTT)检测细胞毒性,槲皮素不同浓度溶液分别作用于被HCMV感染的细胞,通过观察细胞病变效应(CPE),MTT法检测细胞存活率来探讨槲皮素对HCMV感染的细胞治疗作用、对CMV在细胞内增殖的抑制作用及预防作用,从而对其抗病毒机制进行研究。结果:槲皮素的半数中毒浓度(TC50)为4.75mg/ml,对HCMV在细胞内增殖的治疗作用及预防作用的半数有效浓度(EC50)分别为0.30mg/ml和0.25mg/ml,治疗指数(TI)为15.83。结论:槲皮素能明显抑制人巨细胞病毒感染引起的细胞病变效应,CPE抑制率随着药物浓度的增加而增高。Objective:To investigate the anti-cytomegalovirus effect in vitro and mechanism of quercetin from Houttuynia.Methods:Quercetin from Houttuynia was prepared into solutions of different concentrations,MTT assay was used to detect cytotoxicity,quercetin solutions of different concentrations were applied to cytomegalovirus infected cells;the cytological therapeutic effect of quercetin on cytomegalovirus infection,the inhibitive and preventive effect of quercetin on cell proliferation were explored by observing cytopathic effect(CPE)and detecting cell survival rate by MTT method,then the mechanism of anti-cytomegalovirus was investigated.Results:The median toxic concentration of quercetin was 4.75 mg/ml,the median therapeutic and preventive concentrations of quercetin were 0.30 mg/ml and 0.25 mg/ml,respectively,the therapeutic index was 15.83.Conclusion:Quercetin can inhibit CPE induced by human cytomegalovirus infection,the inhibition rate of CPE increases with drug concentration.
