麻疯树脱油种仁的总生物碱:提取及酸性染料比色法含量测定

摘  要:对不同产地麻疯树脱油种仁中的总生物碱进行提取,采用酸性染料比色法,以盐酸川芎嗪为对照品,于420 nm处测定总生物碱含量,并利用单因素法考察最佳提取条件.结果表明,盐酸川芎嗪在0.036~0.180 mg mL-1范围内线性关系良好(R2=0.999 9),平均加样回收率为98.19%,RSD为2.53%(N=6).7个不同产地麻疯树脱油种仁中总生物碱含量在3.817~4.467 mg g-1之间,其中四川攀枝花地区的总生物碱含量最高.该方法操作简便,准确可靠,重复性好,可作为麻疯树脱油种仁总生物碱的含量测定方法.A method was established for extraction and content determination of total alkaloids in defatted kernel of Jatropha curcas,and the contents of the total alkaloids from J.curcas growing in different habitats were compared.With Chuanxiongzine hydrochloridum as control,acid dye colorimetry was used to determine the contents of the total alkaloids at wavelength of 420 nm by UV-visible spectrophotometry,and the optimum extraction conditions were confirmed by single factor tests.The calibration cure of Chuanxiongzine hydrochloridum was in good linearity in the range of 0.036~0.108 mg mL-1(R2=0.999 9),and the average recovery was 98.19%,with RSD of 2.53%(N=6).The contents of the total alkaloids in chloroform extract samples of J.curcas defatted kernel from different origins were 3.817 to 4.467 mg g-1,and the defatted kernel from Panzhihua,Sichuan,China possessed the highest total alkaloids content.This method is simple and accurate with high repeatability,and can be applied to determine total alkaloids in defatted kernel of J.curcas.
