摘 要:目的:探讨健脾理气颗粒对H22肝癌小鼠的抑瘤作用机制。方法:采用细胞接种的方法建立H22肝癌小鼠动物模型,将小鼠分成3组:正常荷瘤组、健脾理气颗粒荷瘤组、健脾理气颗粒脾虚荷瘤组,各10只。检测健脾理气颗粒对H22肝癌小鼠体质量、抑瘤率、血清微量DNA含量、转移指标VEGF表达、凋亡指标Bcl-2表达、诱导分化指标C-myc表达和增值指标PCNA表达的影响。结果:正常荷瘤组小鼠体质量明显减轻,健脾理气颗粒荷瘤组和健脾理气颗粒脾虚荷瘤组小鼠体质量无明显变化,与正常荷瘤组比较具有统计学意义(P<0.05);健脾理气颗粒荷瘤组抑瘤率为28.1%(P<0.05),健脾理气颗粒脾虚荷瘤组为32.4%(P<0.01),具有统计学意义;健脾理气颗粒荷瘤组、健脾理气颗粒脾虚荷瘤组可减少血清微量DNA的表达物,与正常荷瘤组比较,具有统计学意义(P<0.05);健脾理气颗粒荷瘤组、健脾理气颗粒脾虚荷瘤组可降低VEGF的表达,与正常荷瘤组比较,具有统计学意义(P<0.05);健脾理气颗粒脾虚荷瘤组Bcl-2的表达比正常荷瘤组低(P<0.05);健脾理气颗粒荷瘤组、健脾理气颗粒脾虚荷瘤组可下调C-myc的表达,与正常荷瘤组比较,具有统计学意义(P<0.05);正常荷瘤组、健脾理气颗粒荷瘤组、健脾理气颗粒脾虚荷瘤组的PCNA表达无差别。结论:健脾理气颗粒能抑制H22肝癌肿瘤的生长,其作用机制可能是抑制了VEGF的表达,下调了Bcl-2和C-myc的表达,并表现为血清微量DNA含量的下降。Objective: To explore the antitumor effect and its mechanism of Jianpi Liqi Granules on H22 hepatic cancer mice.Methods: After establishing the H22 hepatic cancer mice model by cell vaccination,the cancer-bearing mice were divided into 3 groups: Jianpi Liqi Granules group,Jianpi Liqi Granules Pixu group and control group(regular cancer-bearing mice),with 10 cases in each group.The effect of Jianpi Liqi Granules on the weight of mice,antitumor rate,serum DNA,and the expression of VEGF,Bcl-2,C-myc and PCNA was examined in H22 hepatic cancer mice.Results: A significant weight loss was observed in control group,compared to the Jianpi Liqi Granules group and Jianpi Liqi Granules PiXu group.The tumor-inhibiting rates were 32.4%(P0.01)and 28.1%(P0.05)in Jianpi Liqi Granules PiXu group and Jianpi Liqi Granules group,respectively,which was significantly higher than that in the control group(P0.05).Compared with the control group,lower serum DNA volume,VEGF expression and C-myc expression were observed both in Jianpi Liqi Granules group,Jianpi Liqi Granules PiXu group(P0.05),while the PCNA expression was similar among the three groups.Specifically,lower VEGF expression and Bcl-2 expression were observed in Jianpi Liqi Granules PiXu group(P0.05),compared with the Jianpi Liqi Granules group.Conclusion: Jianpi Liqi Granules can significantly inhibite the growth of H22 hepatic cancer cell in mice.The antitumor mechanism may relate to down-regulating the expressions of VEGF,Bcl-2 and C-myc,while reducing the value of circulating plasma DNA.
