清肺口服液含药血清对小鼠肺上皮细胞TGF-β1及PDGF-BB mRNA基因表达的影响

摘　　要：目的:探讨清肺口服液含药血清对小鼠肺上皮细胞转化生长因子-β1(TGF-β1)、血小板衍生生长因子-BB(PDGF-BB)基因表达的影响。方法:用清肺口服液含药血清、利巴韦林含药血清及空白血清分别作用于体外培养的小鼠肺上皮细胞TC-1,采用实时荧光定量PCR(Real-time PCR)方法测定药物作用前后各组TC-1细胞TGF-β1、PDGF-BB mRNA表达水平。结果:清肺口服液血清作用于TC-1细胞后,使TGF-β1、PDGF-BB mRNA表达水平下调至原来的1/2;利巴韦林血清作用细胞后,细胞中TGF-β1 mRNA升高1倍、PDGF-BB mRNA升高2倍。结论:清肺口服液可能通过抑制TGF-β1与PDGF等细胞因子的过度表达,延缓肺泡炎及肺纤维化形成,在病毒性肺炎的治疗中发挥疗效。Objective： To study the effects of serum containing Qingfei Oral Liquid on expression of transforming growth factor（TGF）-β1,platelet derived growth factor（PDGF）-BB mRNA genes in murine lung epithelial cells.Methods： Intervening in vitro cultured MLE TC-1 cells with serum containing Qingfei Oral Liquid,serum containing Ribavirin and blank serum.Real ¨Ctime fluorescence quantitative PCR method was used to measure the expression level of transforming growth factor（TGF）-β1,platelet derived growth factor（PDGF）-BB mRNA genes.Results： After exposing to serum containing Qingfei Oral Liquid,expression level of transforming growth factor（TGF）-β1,platelet derived growth factor（PDGF）-BB mRNA genes decreased to 1/2;After exposing to serum containing Ribavirin,expression level of TGF-β1 increased to 2 times,PDGF-BB to 3times.Conclusion： Qingfei Oral Liquid may be by inhibiting TGF-β1 and PDGF overexpression of cytokines,slow down the formation of pulmonary alveolus and fibrosis,in the treatment of viral pneumonia to play a curative effect.
