摘 要:目的制备南蛇藤(Celastrus Orbiculatus)乙酸乙酯提取物兔含药血清,研究其对人肝癌SMMC 7721细胞株的增殖、迁移和黏附的影响。方法取新西兰白兔,以20 mg/kg.d南蛇藤提取物灌胃,按血清药理学方法制备含药血清。设低、中、高剂量组(分别含有10%,20%,30%的南蛇藤提取物含药血清),作用人肝癌SMMC 7721细胞,同时设溶媒对照组及5-氟尿嘧啶(5-Fu)阳性对照组。MTT法检测含药血清对细胞增殖的影响,Borden小室体外迁移实验及细胞黏附实验分别检测含药血清对细胞迁移和黏附能力的影响。结果 MTT结果显示各剂量组含药血清均能抑制肝癌SMMC 7721细胞的增殖,且呈剂量依赖性。迁移实验结果表明,中、高剂量组含药血清能显著抑制细胞的迁移能力(P<0.05);黏附实验结果显示在作用1 h和2 h时各剂量组均能显著促进细胞的同质黏附;在作用2 h时,中、高剂量组能抑制细胞与基底膜成分Matrigel的黏附,低、中、高各剂量组均能抑制细胞与人脐静脉内皮细胞的异质黏附,与对照组相比差异显著(P<0.05或P<0.01),并具一定的量效关系。结论南蛇藤提取物含药血清能有效抑制人肝癌SMMC7721细胞的增殖和迁移能力,并能促进细胞的同质黏附能力,抑制细胞与基底膜及与人脐静脉内皮细胞的异质黏附能力。OBJECTIVE This essay aims to prepare rabbit serum containing the Clastrus Orbiculatus extracts(COE) and to investigate preliminarily the effects on proliferation,migration and adhesion in human hepatic carcinoma SMMC 7721 cells.METHODS The New Zealand white rabbits received COE with dose of 20 mg/(kg·d) by gavage.The medicated rabbit serum was obtained with serum phamacological approach.SMMC 7721 cells were cultured in media with different dosages of COE-containing medicated serum(the low,medium,high dose groups containing 10%,20%,30% medicated serum respectively).At the same time,the solvent vehicle control(DMSO) group and 5-Fu positive control group were set up.The proliferation inhibition effect of medicated serum containing COE on SMMC 7721 cells was detected by microculture tetrazolium(MTT) assay.The migration and adhesion ability of SMMC 7721 cells were assayed by Borden chamber experiment and adhesion test.RESULTS COE-containing medicated serum of each dose could significantly inhibit cell proliferation(P〈0.05).Cell migration ability of medium,high dose groups were decreased significantly.After being treated with COE-containing medicated serum,cell homogeneous adhesion ability was significantly promoted,while the heterogenous adhesion ability with Matrigel and human umbilical vein endothelial cell were decreased significantly(P〈0.05,P〈0.01) compared with the negative control group in a dose and time dependent manner.CONCLUSION COE-containing medicated serum can significantly inhibit proliferation,migration and adhesion of SMMC 7721 cells in vitro.
