摘  要:目的:优化DM301大孔吸附树脂纯化补肾方总黄酮的工艺条件及参数。方法:以总黄酮和指标成分含量(二苯乙烯苷、淫羊藿苷和柚皮苷)为指标,采用正交试验方法考察上样量、径高比、洗脱溶剂用量和洗脱速度4个因素对纯化效果的影响,并进行了优化工艺的重复性试验和纯化前后指标成分含量比例变化的考察。结果:经优化的纯化工艺条件为:树脂药材质量比为2∶1,树脂柱径高比为1∶10,洗脱溶剂量为4BV,洗脱速度为2.3BV.h-1,经纯化后总黄酮纯度达50%以上,总黄酮的保留率为76.30%,纯化前后3个指标成分的含量比例基本相同。结论:经该工艺纯化后明显提高了总黄酮的纯度,减少了纯化固形物的量,从而减少服用量。OBJECTIVE To optimize the purification process of total flavones from Bushen Recipe with macroporous resin DM301. METHODS Based on orthogonal design, the contents of total flavones and the contents of stilbeneglycoside,icarrin and naringin worked as indexes, the factors such as aclsorption capacity, the column diameter height ratio, eluant volume and elution velocity were investigated, as well as the changes of the portions of the indicial components before and after purification. RESULTS The optimum elution parameters were as follows: the column diameter-height ratio was 1 : 10, the eluant volume of 70% alcohol was 4 BV and elution velocity was 2. 3 BV·h^-1 , respectively. The purity of the total flavones extract was more than 50%, the reservation rate was 76. 30%, and the yield was 2. 25% (calculating with crude material). The portions of three indicial components before and after purification were nearly same. CONCLUSION The purification process can significantly improve the purity degree and decrease the yield of extract, and thus the administrated amount of the drug was decreased.
